چکیده
Backcrosses of rainbow trout and steelhead (Oncorhynchus mykiss) were used to construct a linkage map and to identify associations between molecular markers and quantitative trait loci (QTL) determining resistance to infectious hematopoietic necrosis virus (IHNV). The sire map covers 1019 centimorgans (cM), and it is composed of 257 molecular markers [185 amplified fragment length polymorphisms (AFLPs) and 72 microsatellites] distributed over 28 linkage groups. The dam map consists of 236 markers (164 AFLPs and 72 microsatellites), with a total of 45 linkage groups, covering 2041 cM. Sixty-one new microsatellite markers were added to the existing rainbow trout linkage maps. Six AFLPs and six microsatellites in the dam map were associated with IHNV resistance and were located in linkage groups 3, 14, 16, 30, 31, and 39. Sixteen AFLPs and six microsatellites associated with IHNV resistance were located in linkage groups 11, 20, and 25 in the sire map. Microsatellite markers mapped to the three linkage groups were used to screen nine other families, and QTL-marker associations were observed for four of the markers in at least one of the families. Further marker mapping is

Genetic variation underlying resistance to infectious hematopoietic necrosis virus in a steelhead trout (Oncorhynchus mykiss) population.
Understanding the mechanisms of host resistance to pathogens will allow insights into the response of wild populations to the emergence of new pathogens. Infectious hematopoietic necrosis virus (IHNV) is endemic to the Pacific Northwest and infectious to Pacific salmon and trout (Oncorhynchus spp.). متن کاملAssociation Mapping of Disease Resistance Traits in Rainbow Trout Using Restriction Site Associated DNA Sequencing
Recent advances in genotyping-by-sequencing have enabled genome-wide association studies in nonmodel species including those in aquaculture programs. As with other aquaculture species, rainbow trout and steelhead (Oncorhynchus mykiss) are susceptible to disease and outbreaks can lead to significant losses. Fish culturists have therefore been pursuing strategies to prevent losses to common patho...
متن کاملMx mRNA expression and RFLP analysis of rainbow trout Oncorhynchus mykiss genetic crosses selected for susceptibility or resistance to IHNV.
Three interferon-inducible Mx genes have been identified in rainbow trout Oncorhynchus mykiss and their roles in virus resistance have yet to be determined. In mice, expression of the Mx1 protein is associated with resistance to influenza virus. We report a study to determine whether there was a correlation between the expression of Mx in rainbow trout and resistance to a fish rhabdovirus, infe...

Single-nucleotide polymorphisms (SNPs) have several advantages over other genetic markers, including lower mutation and genotyping error rates, ease of inter-laboratory standardization, and the prospect of high-throughput, low-cost genotyping. Nevertheless, their development and use has only recently moved beyond model organisms to groups such as salmonid fishes. Oncorhynchus mykiss is a salmon...
